    The effect of Plantago major Linnaeus on serum total sialic acid, lipid-bound sialic acid, some trace elements and minerals after administration of 7,12-dimethylbenz(a)anthracene in rats

    The present study was designed to evaluate the effect of Plantago major Linnaeus (PM) extract on serum total sialic acid (TSA), lipid-bound sialic acid (LSA), some trace elements (copper (Cu), zinc (Zn) and iron) and mineral levels (magnesium, calcium and sodium) in Wistar albino rat administrated 7,12-dimethylbenz(a)anthracene (DMBA). Rats were divided into three equal groups (n = 6). Group I comprised the control group, group II was treated with DMBA (100 mg/kg, single dose) and group III was treated with DMBA (100 mg/kg single dose) and aqueous extract of PM 100 mg/kg/day for 60 days. After 60 days, statistical analyses showed that TSA and LSA levels in DMBA and DMBA + PM groups were significantly higher compared to the control group (TSA: p < 0.01, p < 0.05; LSA: p < 0.05, p < 0.05, respectively). Serum Zn levels were decreased in subjects treated with DMBA (p < 0.01) and DMBA + PM (p < 0.05) compared to the control group values. Serum Cu levels were increased in DMBA group and PM-treated group compared to the control group values. The results of this investigation showed that the levels of TSA and LSA changed significantly, which are sensitive markers for detecting the toxic effects of DMBA. On the other hand, observed decline in Zn levels in rats from DMBA + PM group might be due to decreased generation of free radicals and oxidative stress. Results from this study suggest that PM may be partially effective in preventing carcinogenesis initiated by environmental carcinogen DMBA.

    Fluorescence properties and electrochemical behavior of some schiff bases derived from n-aminopyrimidine

    A series of Schiff bases (L 1, L 2 and L 3 ) were prepared by refluxing aromatic aldehydes with N-Aminopyrimidine derivatives in methanol and ethanol. The structures of synthesized compounds were characterized by FTIR, 1H NMR, 13C NMR and microanalysis. The electrochemical behaviors of the Schiff base ligands were also discussed. Moreover, the evaluation of absorption and emission properties of the structures were carried out in five different solvents. The products show visible absorption maxima in the range of 304-576 nm, and emission maxima from 636 to 736 nm in all solvents tested.

    The changes in the serum, liver, kidney protein and genomic DNA profiles in rats treated with 7,12-Dimethylbenz(A) anthracene and Plantago major L.

    In the study, the changes in the serum, liver, kidney protein and genomic DNA profiles in Wistar albino rats administrated to 7,12-dimethylbenz(a)anthracene (DMBA) and Plantagomajor L. were investigated. SDS-PAGE profiling of the serum proteins showed that the levels of two proteins molecular weighing 140.8 kDa and 46.8 kDa were significantly lower on the 0th day of treatment than on the 60th day of application in the groups. Furthermore, two proteins (34.5 and 22.3 kDa) were solely present in DMBA-treated liver cell lysates. On the contrary, the kidney protein profiles did not show significant banding variations in groups. Genomic DNA analysis also confirmed that DNA extracted from liver and kidney cells did not fragment in any of the groups after the 60th study day. The proteins 34.5 and 22.3 kDa in liver cell lysates at the 60th day in DMBA-treated group could be extra-prognostic indicators for severe DMBA toxicity.

    Silica gel-immobilized 5-aminoisophthalohydrazide: A novel sorbent for solid phase extraction of Cu, Zn and Pb from natural water samples

    A novel silica sorbent, silica gel‐immobilized 5‐aminoisophthalohydrazide (SiO2‐APH), was prepared by the condensation of 3‐chloropropyl‐functionalized silica gel with 5‐aminoisophthalohydrazide (APH) derived from dimethyl 5‐aminoisophthalate as a starting material and used for separation and preconcentration of Cu, Zn, and Pb metals in water samples using Flame Atomic Absorption Spectrometry (FAAS). The characterization of the new sorbent was carried out by Elemental Analysis, Thermogravimetric Analysis (TGA) and Fourier Transform Infrared Spectroscopy (FTIR). Important analytical parameters including as pH, amount of sorbent, type and amount of eluting solvent, sample volume, vortex and ultrasonic bath time, matrix ions that effect the developed SiO2‐APH‐solid phase extraction (SPE) method were investigated and optimum parameters were detected. Recoveries of examined metals were obtained as 98% for Cu and Pb and 101% for Zn. The relative standard deviation (RSD, n = 8) of Cu, Zn and Pb metals were 3.2, 2.8 and 1.6%, respectively. Limit of detections (LODs) (n = 10) were found as 2.7 μg L−1 for Cu, 7.4 μg L−1 for Zn and 3.5 μg L−1 for Pb μg L−1. The accuracy of the new method was assessed by analyzing of TMDA‐51.4 and TMDA‐70.2 certified reference materials. The results obtained for metals were in a good agreement with certified values. Addition/recovery test was applied to the real well, river, dam and stream water samples to check the accuracy of the method. The results showed that the developed SiO2‐APH‐SPE method can be effectively used as an alternative method for determination of Cu, Zn, and Pb metals in water samples.

    Synthesis, characterization and antioxidant activity of heterocyclic Schiff bases

    Schiff base derivatives have gained great importance due to revealing a great number of biological properties. Schiff bases were synthesized by treatment of 4-amino-1,5-dimethyl-2-phenyl-1H-pyrazol-3(2H)-one (1) with various aldehydes in methanol at reflux. In addition, diamine was reacted with an aldehyde to yield the corresponding Schiff bases. The structures of synthesized Schiff bases were elucidated by spectroscopic methods such as microanalysis, 1 H-NMR, 13C-NMR, and FTIR. Antioxidant activities of synthesized Schiff bases were carried out using different antioxidant assays such as 1,1-diphenyl-2-picryl-hydrazyl free radical (DPPH• ) scavenging, 2,20 -azino-bis (3-ethylbenzthiazoline-6-sulfonic acid) (ABTS) radical scavenging, and reducing power activity. (E)-4-((1H-indol-3-yl)methyleneamino)-1,5-dimethyl-2-phenyl-1H-pyrazol-3(2H)-one (3), (E)-1,5-dimethyl-4-((2-methyl-1H-indol-3-yl) methyleneamino)-2-phenyl-1H-pyrazol-3(2H)-one (5), (E)-1,5-dimethyl-2-phenyl-4-(thiophen-2-ylmethyleneamino)-1H-pyrazol-3(2H)-one (7), (E)-1,5-dimethyl2-phenyl-4-(quinolin-2-ylmethyleneamino)-1H-pyrazol-3(2H)-one (9), (1S,2S, N1,N2)-N1,N2-bis((1H-indol-3-yl)methylene)cyclohexane-1,2-diamine (11), and (1S,2S,N1,N2)-N1,N2-bis((2-methyl-1H-indol-3-yl)methylene)cyclohexane-1,2-diamine (12) were synthesized in high yields. Compound 5 displayed a good ABTS•+ activity. Compound 3 revealed the outstanding activity in all assays. Compound 7 has the best-reducing power ability in comparison to other synthesized compounds. Although compounds 5, 11, 12 are new, compounds 3, 7, 9 are known. Due to revealing a good antioxidant activity, the synthesized compounds (3, 5, 7) have the potential to be used as synthetic antioxidant agents.

    Asymmetric organocatalytic efficiency of synthesized chiral β-amino alcohols in ring-opening of glycidol with phenols

    A series of novel chiral β-amino alcohols 3-5 and 7-10 were synthesized by regioselective ring opening of epoxides and chiral amines with a straightforward method in high yields (up to 99 %). Kinetic resolution of racemic glycidol with phenols was achieved by using chiral amino alcohols as organocatalysts. Amino alcohols 5, 8 and 10 exhibited the highest enantioselectivities with p-cresol, phenol, and p-methoxyphenol by 63, 65, 58 % ee, respectively. The moderate enantioselectivities were observed with catalyst 9b towards all the nucleophiles (34-48 % ee). The ee values of the desired 3-aryloxy-1, 2-diols were determined by HPLC. This study presents an attractive tool for the synthesis of β-blockers and structurally complex molecules.

    Voltammetric behavior of testosterone on bismuth film electrode: Highly sensitive determination in pharmaceuticals and human urine by square‐wave adsorptive stripping voltammetry

    In this paper, an electrochemical application of bismuth‐film electrode (BiFE) fabricated via ex‐situ electrodeposition onto a glassy carbon electrode for testosterone determination was investigated in aqueous and aqueous/surfactant solutions. In cyclic voltammetry, the compound showed one irreversible and adsorption‐controlled reduction peak. The BiFE revealed good linear response in the examined concentration range of 1 to 45 nmol L−1 testosterone in BrittonRobinson buffer, pH 5.0 containing 3 mmol L−1 cetyltrimethylammonium bromide. The limit of detection was 0.3 nmol L−1 (0.09 ng mL−1). Finally, the BiFE was satisfactorily applied for quantitation of testosterone in both pharmaceutical (oil‐based ampoule) and biological (human urine) samples.

    Plantago major protective effects on antioxidant status after administration of 7,12-dimethylbenz(a)anthracene in rats

    Aim: The present study was designed to evaluate effects of Plantago major extract on oxidative status in Wistar albino rats administrated 7,12-dimethylbenz(a)anthracene (DMBA). Methods: Rats were divided into three equal groups of 6 animals each: Group 1 controls, group 2 treated with DMBA (100 mg/kg, single dose) and group 3 receiving the DMBA and the aqueous extract at 100 mg/kg/d for 60 days. Results: Significant decrease in catalase (P<0.05), carbonic anhydrase (p≤0.01), reduced glutathione (GSH) (P<0.01) and total protein (P<0.01) values was observed in the DMBA group compared with the healthy controls and DMBA + Plantago major groups. Conclusion: The results suggest preventive effects of Plantago major on DMBA induced oxidative damage in Wistar albino rats that might be due to decreased free radical generation.

    Synthesis, characterization and application of a novel multifunctional stationary phase for hydrophilic interaction/reversed phase mixed-mode chromatography

    A novel multifunctional stationary phase based on silica gel was synthesised starting from L- isoleucine and 4-phenylbutylamine and evaluated as a hydrophilic interaction/reversed-phase mixed-mode stationary phase for high-performance liquid chromatography (HPLC). The prepared stationary phase was characterized by elemental analysis, infrared spectroscopy (IR), scanning electron microscopy (SEM), Brunauer, Emmett and Teller (BET) and solid-state 13C nuclear magnetic resonance (NMR). The mechanisms involved in the chromatographic separation are multi-interaction, including hydrophobic, π-π, hydrogen-bonding, dipole-dipole and ion-dipole interactions. Based on these interactions, successful separation could be achieved among several aromatic compounds having different polarities under both hydrophilic interaction liquid chromatography (HILIC) and reversed phase (RP) condition. Nucleotides/nucleosides were separated in the HILIC mode. The effects of different separation conditions, such as pH value, mobile-phase content, column temperature, buffer concentration and flow rate, on the separation of nucleotides/nucleosides in HILIC mode were investigated. The seven nucleotides/nucleosides were separated within 22 min, while six of them were separated within 10 min by isocratic elution. To determine the influence of the new multifunctional stationary phase under the RP condition, a number of moderately and weakly polar and nonpolar compounds, such as 10 substituted anilines and eight substituted phenols were separated successfully under the RP condition within 14 and 15 min, respectively. Additionally, nine mixtures of polar/nonpolar test compounds were simultaneously separated within 19 min, while seven of them were separated within 12 min, under HILIC/RP mixed-mode conditions. Chromatographic parameters, such as the retention factor and peak asymmetry factor, were calculated for all of the analytes, while the theoretical plate number was calculated for analytes separated by isocratic elution. Compared to traditional C18 and commercial HILIC columns, the new stationary phase exhibited both HILIC and RPLC performance, and the scope of analyte separation was thus enlarged.

    Design, preparation and application of a Pirkle-type chiral stationary phase for enantioseparation of some racemic organic acids and molecular dynamics studies

    This study consists of two parts. In the first part of the study; a Pirkle-type chiral stationary phase was prepared by synthesizing an aromatic amine derivative of (R)-2-amino-1-butanol as a chiral selector and binding to L-tyrosine-modified cyanogen bromide (CNBr)-activated Sepharose 4B and then, packed into the separation column. The chromatographic performance of the separation column was evaluated with racemic mandelic acid and 2-phenylpropionic acid by using phosphate buffers at three different pHs as mobile phase. In the resolution processes, the prepared solutions were loaded onto the separation column at two different concentrations and at three different pHs for each racemic organic acid, separately. Enantiomeric excess (ee%) of the eluates was determined on CHIRALPAK AD-H chiral analytical column by HPLC. The maximum ee% for mandelic acid and 2-phenylpropionic acid was determined to be 60.84 and 27.4, respectively. Separation factors (k1 ’ , k2 ’ , α, and Rs) were calculated for each acid. The structures of the obtained compounds were characterized using the spectroscopic methods (NMR, and elemental analysis). In the second part of the study; enantioselective interactions between the prepared CSP and the analytes have been widely studied by docking, molecular dynamics simulation and quantum mechanical computation methods. The reason of column eluation of rac-2-phenylpropionic acid with lower enantiomeric yield was explained by these techniques.
