    Electroanalytical determination of progesterone hormone using borondoped diamond electrode

    Progesterone is synthesized from pregnenolone, a derivative of cholesterol like other steroids. It is a steroid hormone that has a vital importance in human and animal health. In case of progesterone imbalance, it causes problems such as fertility and infertility. In this study, electrochemical properties of Progesterone hormone Boron doped diamond (BDD) on electrode surface were investigated in strongly acidic environment (HClO4 and H2SO4). A fast, sensitive, selective and simple electroanalytical method was developed for cathodically pre-treated BDD electrodes. A good linear analytical curve in the concentration range of 10 μM to 100 μM at a voltage value of +1.51 V (vs. Ag / AgCl) (after 120 s deposition at +0.1 V voltage) in 0.5 M HClO4 using the square wave stripping technique for the quantification of progesterone hormone obtained. The observability limit is 0.012 µM(3.77 µg L-1); The relative standard deviation value (RSD) for the concentration of 3.1 µM (n = 11) was calculated to be 4.87%. In addition, the developed voltammetric method has been successfully applied to drug and blood serum.

    Bor katkılı elmas elektrot kullanarak progesteron hormonunun elektroanalitik incelenmesi

    Bu tez çalışmasında, Progesteron hormonu Bor katkılı elmas(BDD) elektrot yüzeyinde elektrokimyasal özellikleri, kuvvetli asidik ortamda (HClO4 ve H2SO4) incelenmiştir. Katodik olarak önişlem görmüş BDD elektrot ile hızlı, duyarlı, seçici ve basit bir elektroanalitik yöntem geliştirilmiştir. Kare dalga sıyırma tekniği kullanarak progesteron hormonunun miktar tayinini 0.5 M HClO4 destek elektrolit çözeltisinde +1.51 V (vs. Ag/AgCl) gerilim değerinde (+0.1 V gerilimde 120 s biriktirme sonrası) 0.1-7.1 μM derişim aralığında geniş bir analitik çalışma aralığı elde edilmiştir. Gözlenebilirlik sınırı 0.012 µM (3.77 µg L-1) olup; 3.1 µM (n=11) derişim değeri için bağıl standart sapma değeri (RSD), %4.87 olarak hesaplanmıştır. Geliştirilen voltametrik yöntem ilaç ve kan serum numunelere başarılı bir şekilde uygulanmıştır.

    Method validation and simultaneous determination of retinol, retinyl palmitate, β-Carotene, α-Tocopherol and vitamin C in Rat serum treated with 7,12 dimethylbenz[a]anthracene and plantago major L. By high- Performance liquid chromatography using diode-array detection

    A new and simple high-performance liquid chromatography method was developed and validated for the simultaneous determination of retinol, retinyl palmitate, β-carotene, α-tocopherol and vitamin C in rat serum treated with Plantago Major L. and 7,12 dimethylbenz[a]anthracene. High-performance liquid chromatography analysis was performed utilizing an Inertsil ODS3 reversed phase column with methanol-tetrahydrofuran-water as mobile phase under gradient conditions, at 1.5 mL min-1 flow rate and 25 °C. Diode-array detection was at 325, 450, 290 and 270 nm (retinol and retinyl palmitate), β-carotene, α-tocopherol and vitamin C, respectively and runnig time 18 min. The high-performance liquid chromatography assay and extraction procedure proposed are simple, rapid, sensitive and accurate. The method was then applied for the determination of retinol, retinyl palmitate, β-carotene, α-tocopherol and vitamin C in rat serum. Results of this study demonstrated that; at 60th day DMBA-treated group, there was a significant decrease in vitamin levels compared to the levels of control group. A significant increase was observed in vitamin levels of 7,12 dimethylbenz[a]anthracene+Plantago Major L.-treated group compared to the DMBA-treated group. Additionally, the results obtained in the study are found to be in agreement with data reported in the literature.

    Protective effect of Hypericum perforatum L. on serum and hair trace elements in rats 7,12-dimethylbenz[a]anthracene-induced oxidative stress

    The study was designed to assess the effect of Hypericum perforatum L. (H.P) on serum and hair trace elements and mineral levels, oral administration of 7,12-dimethylbenz[a]anthracene (DMBA) induced oxidative stress in Sprague-Dawley female rats. Analysis of the trace element has been carried out using atomic absorption spectrophotometer method at end of 60th day. It has been found out that the DMBA group contained statistically lower Zn and Cr compared to the control group (p < 0.01) and (p < 0.05), Cu, Mg and Na contained higher than control group (p < 0.05), (p < 0.05) and (p < 0.05). In DMBA + H.P group, Zn higher and Na lower than DMBA group (p < 0.05), (p < 0.05), in hair samples Cd, K and Zn contained lower DMBA compared to the control group (p < 0.05), (p < 0.05) and (p < 0.05). In group DMBA + H.P, Cd was higher than DMBA group and Cr lowered accordance with control group (p < 0.05). The present study demonstrated significantly positive and beneficial effect of H.P on the concentration levels of Zn and Na in serum, also on Cd levels in hair between DMBA and DMBA + H.P groups.
